A PHASE I-II STUDY OF PENTOSTATIN, CYCLOPHOSPHAMIDE, RITUXIMAB, AND MITOXANTRONE IN PREVIOUSLY TREATED PATIENTS WITH CHRONIC LYMPHOCYTIC LEUKEMIA AND OTHER LOW GRADE B-CELL NEOPLASMS
Purpose :

Pentostatin, cyclophosphamide, rituximab, and mitoxantrone are all drugs that are active in chronic lymphocytic leukemia (CLL) and other low-grade B-cell cancers, and have already been approved by the FDA. In this study, investigators want to evaluate the effectiveness of this four-drug combination in patients with these cancers who have had prior treatment for their disease.

Prior studies have shown that the combination of pentostatin, cyclophosphamide, and rituximab can be given safely to patients. In this study, researchers will determine the optimal dose of mitoxantrone that can be given with these three drugs, and they will assess the effectiveness of this four-drug combination in treating these cancers. Laboratory studies suggest that these drugs may be more effective when used together.

Eligibility :

To be eligible for this study, patients must meet several criteria, including but not limited to the following:

  • Patients must have a confirmed diagnosis of CLL or another low-grade B-cell cancer (such as small lymphocytic lymphoma, follicular lymphoma, Waldenstrom's macroglobulinemia, marginal zone lymphoma, or mantle cell lymphoma) that has already been treated with chemotherapy.
  • Patients must be age 18 or older.
