A Phase II Trial of Continuous Low-Dose Temozolomide for Patients with Recurrent Malignant Glioma

A PHASE II TRIAL OF CONTINUOUS LOW-DOSE TEMOZOLOMIDE FOR PATIENTS WITH RECURRENT MALIGNANT GLIOMA
Purpose :

Temozolomide is the most common drug used to treat gliomas. Patients usually take temozolomide for five days once a month. Daily low-dose temozolomide is a different schedule, which may be more effective than the usual schedule because it may overcome some tumor resistance. This schedule may also block the growth of new tumor blood vessels.

This study will evaluate the effectiveness of daily low-dose temozolomide in people with recurrent glioblastoma and other malignant gliomas who have previously taken temozolomide as part of their initial treatment. Researchers will also observe any side effects; see if patients' tumors display evidence of resistance to drugs like temozolomide (measuring a protein called MGMT).; and determine if temozolomide changes the levels of substances that indicate the growth of new blood vessels.

Eligibility :

To be eligible for this study, patients must meet several criteria, including but not limited to the following:

  • Patients must have a glioma that has recurred despite prior treatment with temozolomide.
  • Patients must have recovered from the side effects of previous therapy.
  • Patients must be age 18 or older.
