Summary of Invention
This invention provides streamlined assay methods that rely on fluorescence polarization measurements and/or whole-cell immunodetection of Hsp-90 dependent proteins to identify bioactive compounds that interact with Hsp-90. The first method utilizes minimal amounts of either recombinant Hsp-90 or cellular extracts containing Hsp90. The latter method utilizes a minimal number of cells; yet is sufficiently sensitive and reproducible to permit quantitative determinations.
Advantages
- Maximally efficient identification of Hsp-90 protein inhibitors when both assay methods are used in tandem.
- Ease of use, reproducible, and high sensitivity.
- Suitable for high-throughput screening of compound libraries.
Area of Application
High-throughput screening research tool.
Stage of Development
HSP-90 inhibitor libraries are being screened for potential candidates.
