My research focuses on determining how the anti-tumor immune response becomes inhibited in the tumor microenvironment. Employing intra-vital two photon microscopy, we image deeply into live tumors to study how melanoma tumor-specific CD8+ T cells are rendered anergic. Using fluorescently labeled melanoma tumors, and reporter mice, we measure in real time the interactions between effector CD8+ T cells, regulatory T cell and tumor cells. By studying the outcomes of these interactions, we hope to elucidate the mechanism that prevents melanoma-specific CD8+ T cells from clearing established tumors, in order to design more effective tumor immunotherapy.
