A Phase I Dose Finding Study of Oral LXH254 in Adult Patients with Advanced Solid Tumors Harboring MAPK Pathway Alterations


MAPK is a protein signaling pathway that helps cells grow normally. Alterations in the MAPK pathway can cause cells to grow abnormally and develop into cancer. LXH254 is an investigational drug that works by blocking an abnormal MAPK pathway.

The purpose of this study is to find the highest dose of LXH254 that can be given safely in patients with advanced solid tumors that contain abnormal MAPK. LXH254 is taken orally (by mouth).


To be eligible for this study, patients must meet several criteria, including but not limited to the following:

  • Patients must have an advanced solid tumor that contains at least one MAPK mutation.
  • Patients must recover from the serious side effects of prior therapies before entering the study.
  • Patients must be physically well enough that they are fully ambulatory, capable of all self care, and are capable of all but physically strenuous activities. As an example, patients must be well enough that they would be able to carry out office work or light housework.
  • This study is for patients age 18 and older.
