The role of the Mouse Genetics Core Facility (MGCF) is to support the use of genetically manipulated mice for in vivo studies of gene function in biological processes that are of interest to investigators at Memorial Sloan Kettering Cancer Center. Biological problems investigated using the services and expertise of the MGCF includes cell survival and proliferation, embryonic development and cell differentiation, organogenesis, cell-cell interactions, immune responses, and the generation of mouse models of cancer. The services of the MGCF are provided by the staff of two interacting groups within the Core, the Transgenic Mouse Group and the Colony Management Group.

Services

  • Production of transgenic mice: transgene DNA purification; transgene DNA injection; genotyping of founder mice; and transgene transmission to progeny.

  • Gene Targeting: electroporation of gene targeting vector into ES cells; selection of clones; and identification and cryopreservation of targeted ES cell clones. (This service is offered in conjunction with the facilities and services of the Rockefeller University Gene Targeting Core.)

  • Production of gene targeted mice: generation of chimeric mice by ES cell injection into blastocysts and identification of germ-line chimeras.

  • Cryopreservation of embryos and sperm for long-term storage of transgenic, gene targeted, congenic, and other mutant mouse strains.

  • Rederivation by embryo transfer and in vitro fertilization (IVF) for strain importation and recovery.

  • Performance of specialized animal surgical procedures and embryological techniques based on the needs of investigators.

  • General mouse husbandry procedures for animals housed at Rockefeller Research Laboratories , Zuckerman Research Center, and Long Island City.

  • Management of breeding colonies at Long Island City.

  • Genotyping of transgenic, gene targeted, and mutant mouse lines.

  • Provision of specialized mouse strains/lines for investigators' research.

  • Provision of ES cell services.

  • Consultation & training.
