T-WGBS:
a protocol that utilizes the Tn5 transposome and bisulfite conversion to study 5mC in the genome at single-base resolution. In T-WGBS, a hyperactive Tn5 transposase fragments the DNA and appends sequencing adapters in a single step. T-WGBS requires no more than 20 ng of input DNA; hence, it allows comprehensive methylome analysis of limited amounts of DNA isolated from precious biological specimens. It is a fast protocol with few steps thus preventing loss of DNA.
